Fig. 2. Optimization of conditions for the metalloproteinase-9 (MMP-9) analysis system. (A) NC membrane effect. The ratio of specific signal to background signal was compared simultaneously with the fluorescence signals at different MMP-9 concentrations. In the case of NC #1, although the fluorescence signal was not high, the signal-to-noise ratio was approximately 3.3 times higher than that of NC #2. (B) Sample pad effect. Sample pad #1 is made of glass fiber, while sample pad #2 is made of polyester fiber. The sample pad made of polyester fiber exhibited a signal-to-noise ratio approximately 1.4 times higher. (C) Polyvinyl alcohol (PVA) treatment concentration. The fluorescence signals for polyester fiber at different PVA concentrations were examined. It was found that at lower PVA concentrations, the emission rate decreased, affecting the analysis. Treatment with 0.05% PVA improved the sensitivity.
